煤尘提取物的遗传毒性研究

摘要: 本研究系采集山西省8 个大矿务局所属的10 个煤矿,井下掘进工作面煤样16 份,包括7 种煤、即烟煤、无烟煤、弱粘结煤、肥煤、瘦煤、焦煤、气煤,每一份煤将经提取,分成两份;一份进行亚硝化,另一份为非亚硝化煤样,分别进行Ames 试验,SOS 显色试验,双核微核试验,大鼠骨骼细胞染色体畸变分析试验,结果表明:16 份煤样提取物经亚硝化后,其中有10 种Ames 试验。SOS 显色试验均为阳性,从10 种阳性煤尘中,选1至3 种进行染色体畸变分析及双核微核试验,其结果与Ames 试验,SOS 显色试验一致,阳性率为6215 % ,试验中发现,煤样中挥发分的含量高,亚硝化提取物呈较深的棕色液体,均有致突变性,而随着颜色的加深其致突变性增强,提出煤尘中挥发分含量可间接反映出煤样中有机物的含量,挥发分高的煤样,有机物所占比例较大,经亚硝化后,生成具有致突变作用的亚硝基化合物含量相对高,故可表现出强的致突变性。这一发现,可采纳较为简便的测定挥发分的实验,对煤样致突变性进行初筛,以节省大量的人力物力。

Abstract: In this research ,sixteen coal samples including seven kinds of coals in Shanxi province was investigated ,which including : bituminous coal stome , coal carbonization ,gas coal ,etc. Each coal sample ext ract was divided into two part s : non2nit rosated sample and nit rosated sample.Ames test ,SOS test ,doublenuiclei micronuclei test , rat bone2marrow cell chromosome aberration test indicated :among sixteen coal ext ract s ,ten were positive in Ames test ,SOS test . One or two or three samples f rom the ten positive est ract s were performed by chromosome aberration test and doublenuclei micronuclei test . Positive rate was 62. 5 %. Test s indicate large concent ration of volitile component , nit rosated ext ract s which are heavier brown liquid were mutagenic. The heavier the color was , the more mutagenic the ext ract s were ,which indicated concent ration of volatile component reflected the organic substance content s indirectly. Large concent ration volatile was in large proportion ,which appeared more mutagenic. This discovery provides a much simpler way to test the dispersible content , which can save large amount of human power and material power.
